HyCyte® PBMC-Specific One-Step Cryopreservation Solution (Serum-Free, Non-Programmable)

HyCyte® PBMC-Specific One-Step Cryopreservation Solution (Serum-Free, Non-Programmable) is a rapid, batch-use cryopreservation solution specially developed by Verygenome for PBMC cryopreservation. It is developed specifically for the characteristics of immune cells and NK cells among peripheral blood mononuclear cells (PBMC), reduces cryopreservation-induced damage to NK cells, maintains the proportions of various cell populations unchanged—especially the NK cell recovery rate—and achieves high-survival-rate cryopreservation and recovery of PBMC cells.
The product is easy to use, requiring no special equipment or instruments and no controlled-rate freezing; after resuspending the cells, simply place them directly in a -80°C refrigerator to complete cryopreservation. All components are of high purity with stable chemical properties and are pharmaceutical excipients. The formulation is protein-free, avoiding protein contamination, and is suitable for immune cell and NK cell applications.
1. Collect PBMC cells or isolated buffy coat cells into a centrifuge tube.
2. Calculate the number of cells to be cryopreserved based on the culture cell density and the size of the cryovial used (reference quantity: 5×10^5~1×10^7 cells/mL).
3. Centrifuge to collect the cells. (Reference centrifugation temperature: 4°C).
4. Collect the pelleted culture cells and completely discard the supernatant in the centrifuge tube.
5. Add an appropriate amount of 4°C pre-cooled HyCyte® PBMC-Specific One-Step Cryopreservation Solution to the centrifuge tube. Gently mix the cells to form a cell suspension.
6. Aliquot the cell suspension from the centrifuge tube into labeled cryovials or cryobags; 1 mL or 1.5 mL per vial is recommended, or operate according to cryobag requirements.
7. Immediately place the cryovial directly in a -80°C refrigerator to complete the "rapid" cryopreservation; after 24 hours, transfer to liquid nitrogen or -80°C for long-term storage.
Note: Cryovials or bags should not be placed in close contact; they should be spaced apart to avoid affecting cell cooling.
The product has passed rigorous sterile testing, osmolality, pH inspection, and endotoxin testing to ensure it is free of bacteria, fungi, mycoplasma, viruses, etc.
For routine cell cryopreservation, cell viability is higher than 90%.
