HyCyte® PDO-Specific Cryopreservation Solution
CAT #VG-GUCP-R204-50
HyCyte® Verygenome PDO-Specific Cryopreservation Solution is specially developed for organoid characteristics. It reduces cryopreservation-induced damage to organoids, maintains a high organoid recovery rate, and achieves high-survival-rate organoid cryopreservation and long-term storage for over 10 years.
The product is easy to use, requiring no special equipment or instruments; after resuspending the organoids, perform controlled-rate freezing and place in liquid nitrogen to complete cryopreservation. All components are of high purity with stable chemical properties. The formulation is free of animal proteins and serum, avoiding contamination from animal proteins, making it suitable for organoid applications.
1. When organoid density reaches the cryopreservation requirement, discard the medium, add 1 mL of 4°C pre-cooled passage buffer to each well, gently pipette the matrix gel, and collect the organoids into a 15 mL centrifuge tube.
2. Place the centrifuge tube in a 4°C refrigerator and let stand for 15-30 minutes.
3. Centrifuge at 300 g for 5 minutes and discard the supernatant.
Note: Gently aspirate with a pipette; avoid direct pouring.
4. Add 10 mL of 4°C pre-cooled passage buffer and gently resuspend the organoid pellet.
5. Centrifuge at 300 g for 5 minutes and discard the supernatant.
6. Add 1 mL of organoid cryopreservation solution, gently resuspend, and transfer to a cell cryovial.
Note:
(1) For each 1 mL of cryopreservation solution, the recommended number of organoids to cryopreserve is 1×10^3–1×10^7 organoids of the corresponding cell quantity.
(2) For densely structured organoids, use organoid digestion solution for appropriate digestion, then wash twice with passage buffer before cryopreservation.
7. Place the cryovial in a controlled-rate freezing container (pre-equilibrated to room temperature or 4°C), transfer to a -80°C refrigerator, and after 24 hours transfer to a liquid nitrogen tank for storage.
The product has passed rigorous sterile testing, osmolality, pH inspection, and endotoxin testing to ensure it is free of bacteria, fungi, mycoplasma, and viruses.
Performance testing was conducted using human colorectal cancer organoids, etc.
(1) After resuspending cryopreserved organoids, transfer to the refrigerator for cooling as soon as possible; do not exceed 5 minutes at room temperature.
(2) This product contains DMSO; please wear gloves when handling. Use aseptic technique when using the product to avoid contamination.
(3) For your safety and health, please wear a lab coat and disposable gloves when handling.
(4) This product is for research use only and is not intended for clinical diagnosis or other purposes.
