As an animal-free, ready-to-use recombinant enzyme digestion solution, HyCyte® Animal-Free Gentle Digestion Enzyme is developed with cutting-edge biotechnological engineering and can seamlessly replace traditional porcine or bovine-derived trypsin at a 1:1 ratio without changing existing experimental workflows, easily adapting to various cell culture scenarios. It provides high-viability, high-purity, and high-stability cell samples for downstream critical experiments such as cell sorting, gene editing, and protein expression, making it a core tool in cell culture that combines "gentle protection" with "efficient dissociation." Product Name: HyCyte® GentZyme (Animal-Free Gentle Digestion Enzyme) Catalog No.: GUTP-R005 Storage Conditions: 15–30°C (ambient) / 2–8°C / −5 to −20°C, protect from light Key Advantages ✅ Animal-free safety assurance with more reliable inter-batch stability: Produced via a cGMP-compliant fermentation platform using dedicated animal-free equipment and raw materials throughout, eliminating risks of animal-derived virus and mycoplasma contamination and batch-to-batch variation at the source; rigorous quality testing shows endotoxin < 0.1 EU/mg, host protein residue < 10 PPM, and negative sterility testing, ensuring strong reproducibility for stable support in research and industrial production. ✅ Dual gentle and efficient properties with maximized cell viability: As a high-purity recombinant trypsin analog, it specifically cleaves the C-terminal peptide bonds of lysine and arginine, with a clear mechanism of action as a single enzyme component, avoiding cell damage caused by multiple impurity enzymes in traditional trypsin; while rapidly achieving cell detachment, it perfectly maintains cell surface antigen integrity and membrane structural stability, and for various adherent and strongly adherent cell lines such as 293F, VERO, CHO-K1, A549, and MSC, post-digestion cell viability reaches 95%–100%, significantly outperforming traditional digestion reagents. ✅ Convenient operation that saves time and effort, doubling experimental efficiency: Breaking the low-temperature storage limitation of traditional trypsin, it remains stable stored at room temperature (15–30℃) protected from light, can be stored long-term at 2–8℃ or −5 to −20℃, with a shelf life of up to 24 months, requires no repeated freeze-thaw, and supports flexible experimental scheduling; no inactivation is needed during use, no trypsin inhibitor needs to be added, and the reaction can be terminated simply by dilution with complete medium, simplifying procedures and reducing operational error while greatly saving experimental time. ✅ Broad compatibility across multiple scenarios with flexible application: Compatible with both serum-containing and serum-free culture systems, it efficiently dissociates both routine adherent cells and strongly adherent cells such as PK-15 and MDCK; available in two specifications, 1× ready-to-use and 10× concentrated, with the 10× concentrate flexibly diluted according to cell type to meet different experimental scales and cell characteristics, suitable for research laboratories, biopharmaceuticals, and other scenarios. Key Characteristics Appearance: clear liquid, purity ≥95% by SEC, no degradation or aggregation by SDS-PAGE protein gel Molecular weight: 22.187 kDa, stable enzymatic activity, specific activity ≥27,000 U/mg (at 25℃, pH 7.6, one activity unit defined as the increase of absorbance at 253 nm by 0.003 per minute from BAEE hydrolysis) Physicochemical parameters: osmolarity 275–320 mOsmol/kg, pH 7.00–7.50, highly adapted to the intracellular environment, avoiding cell stress caused by osmotic or pH imbalance Storage & Shipping: safe and stable at room-temperature shipping; storage requires protection from light and avoidance of extreme temperatures Quick Use Guide Preparation: The digestion solution can be used directly at room temperature or pre-warmed to 37℃ to improve digestion efficiency; perform digestion and passage when cells in T25/T75 flasks reach 80–90% confluence. Washing cells: Aspirate the old medium, add 2–3 mL (T25 flask) or 5 mL (T75 flask) of calcium- and magnesium-free PBS, gently wash the cell monolayer twice, and aspirate the PBS. Digestion: Add an appropriate amount of digestion solution (1 mL for T25 flask, 5 mL for T75 flask), ensuring complete coverage of the cell layer, and incubate in a 37℃ incubator for 1–8 minutes (adjust per cell type; use 10× concentrate for strongly adherent cells), observing under the microscope until cells round up and gaps increase. Termination and downstream operations: Immediately add 3–4 mL (T25 flask) or 5–10 mL (T75 flask) of complete medium, gently pipette cells into a single-cell suspension, transfer to a centrifuge tube, and centrifuge at 100×g for 5–10 minutes; aspirate the supernatant, resuspend cells in an appropriate amount of complete medium, and after determining viability with a cell counter, seed for culture or use in downstream experiments. Dilution instructions (10× concentrate): Under sterile conditions, add 1 mL of 100 mM EDTA (pH 8.0) solution to 99 mL of calcium- and magnesium-free PBS to prepare DPBS/1 mM EDTA buffer, then dilute the 10× concentrated digestion solution with this buffer to the desired concentration for use. · For research or further manufacturing use only, not for human diagnosis or direct administration · Wear appropriate protective eyewear, clothing, and gloves during operation to avoid direct contact with skin and mucous membranes · It is recommended to optimize digestion time according to the specific cell line to avoid excessive digestion affecting cell viability
